In today's lab, we performed another ligation reaction with our samples extracted from the gel on 10/20/2011. We had to do another ligation reaction because we accidentally used already ligated samples and re-ligated them. The results of those messed up samples showed some cultures, but they were all blue. It is not surprising the T-vector did not work properly because of our mistake. This time, we took the original sample and set up a ligase reaction. We placed the tubes of our six samples and a positive and negative controls in the freezer to transform into competent E. coli cells in the next lab.
