Finishing Ethanol precipitation and gel electrophoresis

  • We finished the ethanol precipitation and we didn't know if we did it right because we didn't see the pellet of DNA after spinning it in the centrifuge twice. So we went ahead and did an enzyme digest of the precipitaiton and the chloroform extraction and ran it in a gel. We ran two enzyme digested samples of phenol/chloroform extraction and phenol/chloroform with ethanol precipitation in the gel and one undigested sample of each. In our results, we found that the phenol/chloroform with ethanol precipitation yielded better results and we will proceed to PCR with the remaining 9 microliters of DNA.
