* On Tuesday, we ran gel electrophoresis on our now concentrated DNA samples. Only sample 3 (from samples 1, 2, 3, and 5) showed significant protein content. We did not feel confident that this protein product was DNA, so today we ran digested and undigested samples from sample 3 to confirm its DNA content before we proceed. If sample 3 shows that it is liable to digestion, we will proceed with PCR. If not, we will return to our other samples and may also extract DNA from ''Serratia marcescens'' with a new extraction protocol.

Confirmation of DNA content

  • On Tuesday, we ran gel electrophoresis on our now concentrated genomic DNA samples. Only sample 3 (from samples 1, 2, 3, and 5) produced a visible band. We did not feel confident that this was DNA, so today we ran digested and undigested samples from sample 3 to confirm its DNA content before we proceed. If sample 3 shows that it is liable to digestion, we will proceed with PCR. If not, we will return to our other samples and may also extract DNA from Serratia marcescens with a new extraction protocol.
