Samples in lanes 3, 4, and 5 were incubated with Xba1 for 10, 20, and 30min, respectively.  Lane 6 contained undigested plasmid.  Samples in lanes 7, 8, and 9 were incubated with Spe1 for 10, 20, and 30min, respectively.  The gel shows good digestion by both enzymes at all time increments, as compared to circular plasmid, which moved a significantly shorter distance (Lane 6).

Digestion Enzyme Test

  • Tested the digestion of Xba1 and Spe1 enzymes by incubating pSB1A7 plasmid in each enzyme for 10, 20, and 30 min. Results were checked on a gel, compared to undigested plasmid.
