DNA Extract/PCR Prep

  • Chromosomal DNA extracted from Methylosinus trichosporium was pelleted and dissolved in water to be used in PCR reactions to amplify target regions for ligation into pSB1K3 and J62001 plasmid backbones. Working solutions of PCR primers 27F_mmoXY, 27R_mmoXY, 27F_mmoBZDC, and 27R_mmoBZDC (10 um) were made from 100 um stocks.
