Incorporate definitions and examples of general purpose, minimal, selective, differential, enrichment, and defined media in this discussion and include an explanation of how the bacteria that will grow on the particular media that you will use this semester will be selected or differentiated from other microorganisms that are likely to be found in your particular soil community. This discussion will require some outside research into culture media, soil community microorganisms in general, and those microorganism more specific to your habitat or particular eco-system or rhizosphere. Since outside sources of information that is discovered through research must give credit to the investigators who published cited findings, you will need to include a References page. In BISC209 we will use the Name/Year citation style of the journal ''Cell''.

6 7 Write a brief summary of the theory behind the following techniques that we used to identify our bacterial species by molecular tools: genomic DNA isolation, polymerase chain amplification of part of the 16s rRNA genes, use of the Zero Blunt® TOPO® PCR Cloning Kit to create a library of unique plasmid vector with our 16S rRNA gene inserts and then select, One Shot® TOP10 Competent E. coli Cells that allowed us to select and separate our 16S rRNA genes for sequencing, and DNA sequencing by the Sanger method. 5
