In order to maintain the utility of our GAIIs, the BioMicro Center has developed methods to run the GA in a lane-wise manner. These methods are not supported by Illumina and we are learning by trial and error. Below are some validation experiments we have done:
Testing lanewise flow | Food coloring was loaded in the GAII using one of the standard 50ml conicals and flowed through a used flowcell either 1 lane at a time or 4 lanes at a time. | |
Testing reclustering of the flowcell | A single flowcell was subjected to 5 cycles of clustering and sequencing over ~2 weeks and evaluated for various quality parameters. PhiX was used as the test sample. Sequencing reagents were replaced for the 4th sequencing cycle. | |
