The BMC is currently testing the Beckman SPRIworks library preparation system. This platform is a fully automated library preparation system for Illumina sequencing and supports all Illumina applications except for smallRNA-Seq. With the exception of Enrichment, it performs all of the traditional Illumina protocol steps listed on the Illumina Library Preparation page. A total of 10 libraries may be simultaneously prepared in 5 hours for Illumina sequencing.

How it Works

The SPRI-works system consists of three main components: the SPRI-TE Nucleic Acid Extractor, a method card that controls liquid handling, and a cartridge containing all of the reagents required to prepare a single library.

This platform utilizes AMPureXP DNA binding magnetic beads to purify fragmented DNA and perform size selection, replacing the need for column purification and gel-based size selection. There are two size range options available, 200-400bp or 300-600bp. There is also the choice to perform no size selection. After the size selection and adapter ligation process is complete on the SPRI-TE, samples will be ready for PCR Enrichment and clean-up to complete the library prep process.

Sample Submission

Sample concentrations for the SPRI-TE robot are highly variable, we have successfully run samples as low as 0.3ng/uL. It is suggested that you submit as much volume as possible, as the first step of the protocol is to bring all sample volumes up to 100uLs in water.

All samples submitted for direct SPRI-Works library preparation must be submitted with user supplied adapter mix. Adapter mixes submitted should be previously diluted and should have enough volume for 10uLs of adapter per sample. For recommended dilutions for various sample concentrations please see the chart below:

Please note that samples will need to be resubmitted separately for Illumina sequencing.
