Rhodopsin Amplification Test

  • This test was used to assess whether or not Rhodopsin PCR can be used to distinguish between samples that genotyped well, vs those that didn't.
  • In this first test done on August 01 2017, 1.5 μL of YWAR DNA was used with 40 amplification cycles. The first row is of samples that did genotype well and the second row are the ones that didn't genotype well. All samples amplified and our results were inconclusive.

  • Second test was done August 03 2017. Only 1 μL of YWAR DNA was used on the same samples as above and with only 20 amp cycles-- done in hopes of getting a lighter gel band to see if we can distinguish between the two sets of samples based on band brightness. The result was that nothing amplified.

  • Aug 09 2017 third test used 1 μL of DNA with 30 PCR cycles, first 4 samples 13081-13084(Blood Extracts) all SNP genotyped well. Samples 11011-11014 didn't genotype well showed lighter bands.

  • The next objective would be to try feathers that genotyped well vs those that didn't. Also would be good to include the blood samples again to compare brightness.
