On Thursday 24th, we will continue with the cloning of GA20ox. You will run your digested PCR product and vector on a gel, purify your DNA and set up the ligation.

Read page 7 of the GA5_Complementation lab manual (until step 10). Please have your summary ready for that. We will also finish our TAIR worksheet.
Next Tuesday you will transform your ligation into E. coli cells. So please write up your summary and your protocol for that section (until step 12).

We will also discuss the paper Spielmeyer et al. (2002). Please read it and complete the worksheet as homework for Tuesday. We will discuss your conclussions in class as well. I have posted the paper and the worksheet on Angel.

On Tuesday Aprril 5th (in two weeks) I  will collect your labbooks for grading, they should include all the details of every experiment you have done so far, your results (for example the image of your gel and the data on your GA4 spraying experiment) and any comments/changes we discussed in class. Please talk to me if you are not sure what should go in there.
We will also finish our TAIR worksheet.
Please also remember to check your schedules for the final exam.
