Inoculation of GFP/ATF1 Recombinant Colonies

  • Today we inoculated - (continued growing) - the two colonies discovered on Friday. (They had not increased since then, so we are skeptical.) We pipetted 5mL of LB solution into 10mL tubes, added 5uL of ampicillin, and used pipette tips to scrape a bit out of each colony. The tips were dropped into the LB solution/ampicillin and the 10mL tubes were labeled and put into the shaker at 37-degrees Celsius overnight. The plates were kept at room temperature overnight.
