Abstract

Measure thickness and lateral size of sample with the inverted microscope

Equipment

Microscope : Olympus BX41 Microscope camera : Cool snap HQ2

Procedure

For thickness (Z):

- Focus on top of your sample.
- Read the graduation on the fine adjustment knob.
- Focus on the bottom of your sample.
- Read the graduation on the fine adjustment knob.
- Subtract two readings.
- Multiply this number by 1.2 to have your thickness in micrometer approximately.

For lateral size (X,Y):

- Take a picture of your sample.
- For 10x50x → 1pixel≈129.7μm
- For 10x20x → 1pixel≈325.7μm
