Procedure
- Resuspend pellet in a mixture of 45 μL ddH2O and 5 μL NEB DNase buffer
- Add 1 μL of DNAse I (in freezer; keep on ice)
- Mix thoroughly
- Place in 37°C water bath for 10 minutes
- Add 2 μL of 0.25 M EDTA
- Heat at 75°C for 10 minutes
- Store sample on ice or in -20°C freezer.
When ready proceed to Reverse transcription
