Poured large gel with tiny combs. Ran dilutions of ladder and PCR products from last night. Some of them worked, some didn't. Possible the DMSO is messing things up.



PCR gradient with no DMSO of a sample that didn't have a band on the last gel. Gradient from 66-56°C. Didn't see any bands at all.

Samples from gRNA 20 all had bands. Can at least test those. Should redo the PCR though. Will do one more test PCR tonight and then do gRNA 20 samples tomorrow.

Test: run a gradient PCR of a sample that I've had success with before with and without DMSO.
