Based on the results of the gel we ran, there is reason to believe that the ligation worked in some of the colonies. So, I picked colonies 6, 8, 9, and 10. 6 and 8 are from the 3:1 plate and 9 and 10 are from the 1:3 plate. I put them in liquid cultures and placed them in the shaker overnight to grow.
