Summary

  • Liquid Cultures

Picking Colonies; Liquid Cultures
No growth on any LCR plates (likely due to linearized vector dilution or old parts).
~100 colonies on successfully constructed PcTF (BL09, BL01) and BL05 in V0120 retransformation plates.
No growth on either negative (new amp seems to have fixed this).
Two colonies picked from each retransformation plate.
Grew up clones in 4mL LB amp media in shaker overnight.
