The following parts are Amp resistant and from the bacteria, Pantoea ananatis. The numbers correspond to the labeling used in the lab:

I pipetted 10ul of water into each well in the kit plates and transfered the DNA to 1.5ml tubes. I added 30ul of thawed DH5alpha competent cells to each tube, gently mixed, and left the tubes on ice for 5 minutes. I then pipetted the cells onto LB+amp plates and left overnight at 36deg (maybe 30? check warm room temp).
