Today's Procedure and Calculations

Making Hydrogels:

2 hydrogels of 22k MW PVOH and 2 hydrogels of 146-186k MW PVOH. All will have 1% isopropanol: PVOH OH groups. 1 of each will have glutaraldehyde (1mL).

From previous calculations, 1g PVOH contains 1.373E22 OH groups. We want 1% of these to react w isopropanol at 1:1. (1.373E22)(0.01)=1.373E20 molecules of isopropanol. (1.373E20)/(6.022E23 molecules per mole)=2.27997E-4 moles isopropanol. (2.27994E-4 moles)(60.1 grams per mole)=0.0137 grams isopropanol. (0.0137g)/(0.786 grams per mL)=0.0174mL Isopropanol.

1) 1g 22000mw PVOH dissolved in 20mL distilled water + 0.0174mL Isopropanol once PVOH fully dissolved. Poured into dish, sealed with parafilm, and placed in freezer.

2) 1g 22000mw PVOH dissolved in 20mL distilled water + 0.0174mL Isopropanol and 1mL glutaraldehyde (in hood) once PVOH fully dissolved. Poured into dish, sealed with parafilm, and placed in freezer.

3) 1g 146-186K mw PVOH dissolved in 30mL distilled water + 0.0174mL Isopropanol once PVOH fully dissolved. Poured into dish, sealed with parafilm, and placed in freezer.

4) 1g 146-186K mw PVOH dissolved in 30mL distilled water + 0.0174mL Isopropanol and 1mL glutaraldehyde (in hood) once PVOH fully dissolved. Poured into dish, sealed with parafilm, and placed in freezer.

crosslinking couldn't occur bc we didn't have time. To be done next time.

Notes

  • the 148-186K films never really fully dissolved
  • I accidentally added glutaraldehyde to both of the 146-186Ks
