  • Did transformation of the ligations from yesterday of LacI with RBS-Tes with both negative and positive control.

ETHANOL:

  • Started shake flasks of seeds started yesterday.
    • Took 500 μL cells, added to new 50 ml LB+ 50 μL chloramphenicol
    • Waited till O.D. reached .7, added 25 μL IPTG (.5 mM) (about 2:00 PM)
    • Let grow overnight
