TesA+RBS:

  • Transformed ligation/- control that was in freezer.
  • +Control: mCherry 3 μL
  • 200 μL of mixture on chlor and 200 on amp.

ACC Plasmid:

  • Ran a gel of amplified plasmid parts to ensure that the PCR worked.
-Gel looked good, but was very faint. bands were in the proper places.

Ethanol:

  • Re-transformed PACYC plasmid into BL21 cell line.
