Prepare chromosomal DNA extraction from Bacillus

- Grow 2 tubes into LB for

  • IA751
  • IA751 + ECE153 from 03/09 (for tissue kit)
  • IA751 + ECE153 from 02/09
  • IA751 + ECE112 from 05/09

PCR AmyE from ECE112

- Add 12μL of SDW, 5 μL of MM, 1μL + 1μL of each primers (for AmyE back and AmyE front) and 1μL of ECE112

  • Lane1 : HyperladderI
  • Lane4 : AmyE front
  • Lane5 : AmyE front
  • Lane6 : AmyE back
  • Lane7 : AmyE back
  • Results : good band for AmyE front, very low band for AmyE back
