The mutants were streaked out from the innoculation discs onto both kanamycin and non antibiotic plates for the resistant strains, and just onto the normal plates for non-resistant strains. They were then incubated overnight at 37°C. The flame photometer was also calibrated using μM concentrations of potassium.
