To-do
- Inoculate transformants of KaiA + bkb and KaiB + bkb ligations
- Inoculate Top10F' transformants (J04450 plasmid)
- Run the X-P digest of J04450 on a gel, extract and purify the J04450 insert if it exists
- Miniprep the J04450 in Top10 culture that we inoculated last night
- (Maybe) Perform another genomic PCR extraction of KaiA/B and KaiC
Top10F' transformants are constitutively red
It looks like the Top10F' cells we transformed with J04450 are constitutively expressing RFP, despite the lacIq gene on the Top10F' genome.
