20ul digestion

5~8ug  DNA (inserts, if vector, max of 0.5ug) 
2ul  10x buffer

2ul Enzyme – add last (must be kept cold at all times) (should be less than 10% of the total volume otherwise glycerol will inhibit the enzyme activity) 

Make it up to 20ul with MiliQ 
1hr at 37oC.
