*To mimic Lotka-Volterra, the predator construct is split into a 'sensing part' and 'killing part'.
*To measure the Km and Vm of LuxR in our cells.
*The sensing part of the predator produces AHL lactonase as a function of the AHL produced by the prey

*In this part the AiiA gene has been replaced with the reporter gene GFP to allow the measurement of the production of GFP as a function of AHL.

*The resulting transfer curve will be used to extract parameters Vmax and Kd to characterize the part and for use in our modelling.

*This part will form the receiver of the final predator construct
This expeiment will characterise the transfer function of the parts J37016 and J37020. One of these parts will form the reciver in the predator cell. There are many similarities between this characterisation and that of the prey reciver

Motivation

  • To mimic Lotka-Volterra, the predator construct is split into a 'sensing part' and 'killing part'.
  • The sensing part of the predator produces AHL lactonase as a function of the AHL produced by the prey
  • In this part the AiiA gene has been replaced with the reporter gene GFP to allow the measurement of the production of GFP as a function of AHL.
  • The resulting transfer curve will be used to extract parameters Vmax and Kd to characterize the part and for use in our modelling.
  • This part will form the receiver of the final predator construct
