Miniprep of the O/N culture of pSC008

I use the thermoscientific kit, but with the vacuum method. After Centrifugation, resuspension, lysis, neutralization, centrifugation for 5', I plug the column on the vacuum machin, and then add the supernatant, switch on the vacuum, let the liquid pass through. Once there is no more liquid, I add 700μL of wash solution, twice. I centrifuge 1' to remove ethanol, and I finally elute in 50μL of water.

  • Nanodrop : 301 ng/μL : works well

    • DNA : 45 µL / 3 / 3
    • EcoRI : 14 µL / 1 / 0
    • PstI : 14 µL / 0 / 0
    • buf. 10X : 28 µL / 2 / 2
    • water : 179 µL / 14 / 15
      • Final Volume : 140µL/20/20

--> 120 min, 37°C water bath.
