From the first round of sequencing, only 2 over 6 have not the good mutation, and these 2 are those who didn't grow on Kan, because they have frame-shift or lots of mutation. | |||
It turns out that XL1 blue cells, NEB turbo, DH5α have all an amber suppressor in their genome (supE44, or glnV44). I re-transform 2µL in 20µL of competent MG1655 from Jake's stock, and I plate 200µL on LBA+Amp. | |||
