Preparation of next week

  • I made a transformation of the BB with the backbone pSB1T3. I transformed 2µL in 20µL of cell.
  • I recover from -80°C stock :
    • pSB4K5 for the QCM quick change
    • pSC013/014 to make electro-competent cell tomorrow.
  • The transformation worked. Control are as expected.
  • I took 2 clones from the morning transformation, and I make them grow in LB + Tet, for further miniprep.
