What we did today

  • Checked the plates of the ligation product+transformation
    • both negative controls have nothing
    • one (two?) colonies on pSG.005 plate
    • no colony on pSG.006 plate
    • lots of colonies on positive control
  • Cultured pSG.005 from the plate in LB+Cm
  • Ligated (after digestion with X and P):
    • pSG.001 as negative control
    • pSG.002 as negative control
    • pSG.005 (pSG.001+pRG.006 (pLac from Antoine digested 11/07/12))
    • psG.006 (pSG.002+pSG.004 (pTet digested 11/07/12))
  • Transformed the ligation product and plated (plus pSG.002 as positive control)
