PCR wasn't successful for the Pfu crp* runs so ran another PCR with new thermocycler protocol:

  • Cycle 1:(1x)
    • Step 1: 95C for 1:00
  • Cycle 2:(28x)
    • Step 1: 95C for 0:15
    • Step 2: 55C for 0:30
    • Step 3: 72C-75C for 0:45
    • Gradient: H=72C . A=75C
  • Cycle 3:(1x) Hold at 4C


9:30-6
