August 21, 2007

  • Set up 20uL double digests with psB1A2 as well as sequential digests with EcoR1 and SpeI
  • Used MP from galen's box marked:
    • 6B for E+S
    • 4B for E
    • 3B for S
  • Started incubating at 11:03am
  • Ran gel on restriction products
  • Started @ 1:50pm
  • Started an overnight ligation at 16C with old crp* 4uL restriction E+S from yesterday and the gel extracted psB1A2 restriction E+S from 8/10
  • Used a 6:2 ins:vect ratio
