- Weekend plan:
- Friday evening: Transform into DH5a, plate on LB Amp, Also innoculate culture of DH5a
- Saturday: Innoculate cultures from plates if transformation is successful.
- Saturday: DH5a competent cell prep if unsuccessful, followed by transformation.
- Transformed crp* into DH5a. Only had enough competent cells for 6 transformations
- Transformed Ligation #: 1,4,6,7,9,10
- Plated on LB Amp and put in 47C incubator overnight
- 10:30-12:30am
- 3:30-6pm
