September 7, 2007

  • Weekend plan:
    • Friday evening: Transform into DH5a, plate on LB Amp, Also innoculate culture of DH5a
    • Saturday: Innoculate cultures from plates if transformation is successful.
    • Saturday: DH5a competent cell prep if unsuccessful, followed by transformation.
  • Transformed crp* into DH5a. Only had enough competent cells for 6 transformations
    • Transformed Ligation #: 1,4,6,7,9,10
    • Plated on LB Amp and put in 47C incubator overnight


  • 10:30-12:30am
  • 3:30-6pm
