August 13th, 2009measured DNA conc. using nanodrop appartus. this DNA was cut using PacI and AscI (overnight) and then gel purified. Ligation (super GFP into Longtine Plasmids [Kan and His]) 20 ul RXNs 4 5 x buffer 2. plasmid 2. insert 1 igase 11 ddH2O Ran four reactions 1. S-GFP + His Plasmid 2. S-GRFP + kan plasmid
