June 25, 2010

  • Prepared LB agar and made assorted antibiotic plates (LB+ampicilin, LB+kanamycin, LB+ampicilin and kanamycin) for co-transformation
  • Received ethanol and kanamycin from Julie
  • Cleaned the lab
  • Prepared new mini prep buffers from new mini prep kit
  • Mini prepped CFP, two of 1.5 ml of cells and two of 1.0 ml of cells
  • Co-transformed TOP10 two plasmids (PUC 19 and PSB2K3)
  • Sara and Daniel met with Matt Lux to discuss bio brick construction, optimization of fluorescent proteins, and spectral un mixing
  • Amanda, Dasha, and Joe discussed primer sequences with Matt Lux
