Golden Gate Ligations
Follow Nicola Patron's startup guide. If there are internal BsaI or BpiI sites add these additional steps to the end:
1) Add 1.5 μL T4 ligase buffer and 0.5 μL T4 Ligase
2) Incubate at 16°C for 1 hour
3) Heat to 65°C for 10 min
4) Transform into Top10 cells
