Fig. 1. The design of DNA origami “The Tortoise, the Hare and the ground.” This structure is called T1H6, which denotes the Tortoise at position 1 and the Hare at position 6. We designed this whole structure by hands, without using caDNAno at all.

DNA origami “The Tortoise and the Hare (T1H6)” was clearly of observed by using AFM (Fig. 2.) The Tortoise is located on the left side and the Hare is located on the right of the ground. But in the AFM image, we also observed two motifs joined to each other. We solved this problem by adding T4 loops to the staples. But this was not enough. We assumed that single-stranded Loops are also responsible for aggregation. So we added short strands which are partially complementarity to the Loops (Fig. 1.)









We could observe another five frames.
