We succeeded to observe DNA origami “The Tortoise and the Hare” by using AFM. (Fig. 2.) The Tortoise is located on the left side and the Hare is located on the right of the ground. But in the AFM image, we also observed two motifs joined to each other (Fig. 3.) We solved this problem by adding free Ts to the staples and staples which are complementarity to loops to prevent Linkers from stacking.(Fig. 1.) We will make the Trees with staples on the Ground. If we observed AFM images "the Trees", we will connect Two AFM images "The Tortoise, the Hare and the Ground” and "the Trees".(Fig. 6.)
