Our lab ~~is investigating~~ the mechanisms by which extracellular oxidation (by inflammation), and intracellular oxidation (such as initiated by receptor ligation) influence the ability of cells to signal. We rely upon a strong synergy between computational and experimental methods to characterize proteomic dynamics of thiol oxidation. Because of the numerous biochemical reactions involved, we use computational modeling to investigate how signaling networks are regulated ~~by redox-sensitive proteins, whose activities and/or functionality change~~ in the presence of reactive oxygen species. Experimentally, we are developing novel high-throughput ~~biochemical assays to detect~~ and ~~quantify~~ reversible ~~cysteine~~ oxidation ~~of proteins~~.	+	How intracellular and extracellular environments control the transmission of cellular information is important for our understanding of cellular function. Our lab investigates the mechanisms by which extracellular oxidation (by inflammation), and intracellular oxidation (such as initiated by receptor ligation) influence the ability of cells to signal. We rely upon a strong synergy between computational and experimental methods to characterize proteomic dynamics of thiol oxidation. Because of the numerous biochemical reactions involved, we use computational modeling to investigate how signaling networks are regulated in the presence of reactive oxygen species by changes in activity and/or function of redox-sensitive proteins. Experimentally, we are developing novel high-throughput techniques for the detection and quantification of reversible protein oxidation.
