1:00pm Malisha, Fabiana and Reuben then looked at the slides from embryos RC002 and RC003 stained by Reuben under a microscope. We have decided to stick with nissl staining and leave H&E staining.

9:00am Following emailing my notes on cryostat sectioning to Satya she said she would grant me permission to publish the document to OpenWetWare. After making some updates and revisions and spell-checks I have emailed the document back to Satya to get clearance to publish it. --Reuben Cutfield 15:28, 9 December 2010 (EST)

9:50am I emailed Satya our H&E protocol as well. If she clears it too I will also publish it to OpenWetWare. --Reuben Cutfield 15:51, 9 December 2010 (EST)

10:30am I had a meeting with Satya and discussed the changes to the document. We went through it together and made some revisions. Once I have made the changes completely I will publish it online. --Reuben Cutfield 17:57, 9 December 2010 (EST)

11:00am I stained the sections 1,3,5,7 from embryo RC003 cut by Fabiana yesterday using the H&E stain. I varied the time spent in alcohol, haematoxylin and eosin and recorded the times in my lab book. They are now cover-slipped and ready to be viewed. --Reuben Cutfield 17:57, 9 December 2010 (EST)

12:00 Performed nissl stains on the remaining slides 2,4,6. Leaving in nissl for 2,4 and 6 minutes respectfully.

1:30pm I made the final revision to my document about cryomicrotomy and published it to the protocols page --Reuben Cutfield 20:24, 9 December 2010 (EST)

Malisha

Fabiana cut embryo MH003 Results from cutting exercise:

There is still damage evident however there is less damage to the tissue compared to when it was frozen. The conclusion was to cut below a temperature of -27 OT/CT and at 25 microns--Malisha Hettiarachchi 20:26, 12 December 2010 (EST)
