Rich Media for Caulobacter crescentus
Ingredients
- 2.0g bactopeptone
- 1.0g yeast extract
- 0.3g MgSO4
- From Sigma
- 10mL 50 mM CaCl2
- From Emanuele: not necessary for Caulobacter growth at MIT (perhaps enough calcium already in yeast extract or kitchen flasks).
- Now we use 1mL of 0.5M CaCl2 from the MIT Kitchen --Sri Kosuri (talk) 16:49, 19 April 2007 (EDT)
- 1L H2O (milliQ or distilled)
Instructions
- Add ingredients and autoclave (250°F, 22psi, 30-60min depending on volume).
- For plates, usually add 1.5% (w/v) BactoAgar as well.
Notes
- If PYE is to be aliquoted, first prepare 1L as described, then aliquot/pour into 100mL bottles. Finally, reautoclave individual bottles
