Rich Media for Caulobacter crescentus

Ingredients

  • 2.0g bactopeptone
  • 1.0g yeast extract
  • 0.3g MgSO4
    • From Sigma
  • 10mL 50 mM CaCl2
    • From Emanuele: not necessary for Caulobacter growth at MIT (perhaps enough calcium already in yeast extract or kitchen flasks).

  • 1L H2O (milliQ or distilled)

Instructions

  • Add ingredients and autoclave (250°F, 22psi, 30-60min depending on volume).
  • For plates, usually add 1.5% (w/v) BactoAgar as well.

Notes

  • If PYE is to be aliquoted, first prepare 1L as described, then aliquot/pour into 100mL bottles. Finally, reautoclave individual bottles
