HOW TO ... MAKE PCR REAGENTS

1- 5x Tris Boric acid EDTA buffer

For 2 liters: - TRIS base (Boehringer Mannheim 1814273): 108 g - Boric acid electrophoresis grade (FisherBiotech BP168-500): 55 g - 0.5 M EDTA (pH 8.0): 40 ml (or 9.3 g Na2EDTA) - adjust to pH 8.3 - H2O to 2 liters


2- EDTA 0.5 M pH = 8.0 - EDTA: 18.61 g - NaOH: 2.0 g - H2O: 80 ml - dissolve on a magnetic stirrer - adjust to pH = 8.0 with NaOH 10N and 1N - H2O to 100 ml
