McClean:General Flow Cytometry Procedure

Materials

1. PBS + 0.1% (vol/vol) Tween 20
2. Falcon 2058 6mL tubes
3. a sonicator
4. BD LSRII

Procedure

1. For each sample fill a falcon tube with 0.8mL of PBS + 0.1% Tween and put it on ice
2. Grow yeast up and run experiment
3. When you want to take a sample pipet 250 μL of culture into the corresponding falcon tube
4. Sonicate the cells with ten 0.5 second long bursts.
5. Measure fluorescence with the BD LSRII
