Overview

Yeast nitrogen base without amino acids with agar for plates. This protocol leaves 100ml (per L) of room for additives (glucose, amino acids, etc). Media is labelled as YNB1 on the media shelf.

Reagents

  • Yeast Nitrogen Base (without amino acids)
  • Bacto Agar
  • MilliQ H2O

Protocol

  • Container 1:
    • Begin with approx. 700ml MilliQ H2O in 2L bottle
    • Add with constant stirring: 20g of Bacto Agar
    • Bring up to 800ml with MilliQ H2O
    • Autoclave
    • Allow to cool (50°F)
  • Container 2:
    • Begin with approx. 80ml MilliQ H2O
    • Add with constant stirring 6.7g of Yeast Nitrogen base (without amino acids)
    • Bring up to 100ml with MilliQ H2O
    • Autoclave
    • Allow to cool (50°F)

Combine contents of 2 containers and mix well
