This recipe makes 1L of media supplemented with

  • xxx% of selected carbon source
  • 1 mM thiamine hydrochloride
  • 0.2% casamino acids

Combine all solutions using sterile technique:

  • 200 mL 5x M9 salts
  • 30 mL of 10mg/mL thaimine hydrochlorite
    1. Dissolve 10mg per mL of H2O
    2. Filter-sterilize using a 0.22μm filter
    3. Light-sensitive. Store covered in foil
  • 20 mL of 20% carbon source
  • 20 mL of 10% casamino acids
  • 20 mL of 0.1 M MgSO4
  • 1 mL of 0.1 M CaCl2
  • deionized water to 1L (709.8mL)

Store at 4°C.

Note: Adding Fe increases cell density.
