• transform your mutant into Rosetta2 cells
• transform TFAM into Rosetta2 cells
• pour 1 or 2 SDS-PAGE gels
• prepare DEAE cellulose according to instructions (look for it on protocols page)

• make 1 L of 1 M Tris-HCl, pH 8.0

• wash out 2 L flasks and make 6L of LB and autoclave

• make tet-LB plates
  
• streak Xl1-blue onto tet-LB plate
  
• prepare top-agar
