• make competent Xl1-blue cells
• transform dsM13 into Xl1-blue
• pour two SDS-PAGE gels (12%)
• run fractions from the polg-b mutant on SDS-PAGE
• spin down the cells in the shaker
• make 6 L of LB and autoclave, plus 4 - 100 ml flasks
• start overnight culture of RNAp (AMP/Chloro)
