PBS stands for phosphate buffered saline. Adding of the detergent Tween 20 or Triton X-100 makes it PBST (people use the name PBST/PBS-T for both).

Purpose

  • PBST (Triton) is for example used for washing in immunohistology. A 1% PBS Triton X-100 wash can also be added to fixed tissue sections for 10 minutes followed by 3 x 15 minutes PBS wash to permeabilize tissue prior to blocking at the beginning of a immunostaining experiment.
  • PBST is used in protein blots for washing of membrane labelled with antibody.

Recipe

0.1% (v/v) Tween 20 in 1x PBS

examples

  • 100µl in 100ml 1x PBS
  • 1ml in 1L 1xPBS
