  • Excess iodoacetamide can cause primary amine N-terminal carbamidomethylation of peptides. see here
  • Iodoacetamide can cause conflict with Ubiquitin GG patterns, but this side-effect is eliminated when the reaction is performed at 21 °C or cooler. (article | relevant supplementary data)
  • It is thought that formic acid in acetonitrile causes polymerization and contribute to plugging of tips and the HPLC. One can prevent this by making fresh ACN/formic acid mixes weekly, or just use straight ACN (no formic acid). The water and formic acid is OK. [Reference to the ABRF discussion on this??]
