(10 mg/ml)

  • ssDNA is stored in cold box (dessicated)
  • weigh out enough for 10 mg/ml in dH2O
  • stir at room temperature until dissolved (takes a long time)
  • shear DNA by passing 6 times through an 18-gauge needle and 6 times through a 22-gauge needle
  • bring up to 3x volume with dH20, and then boil solution until it is back at 1x volume.
  • aliquot into 1.5 ml Eppendorf tubes and store at –20 °C.
